Parkinson's disease (PD) is a chronic neurodegenerative disease that affects the nigrostriatal dopaminergic complex, causing a variety of motor and non-motor symptoms. One of the main forms of treatment for the disease in its advanced stage is neurosurgery for deep brain stimulation (DBS), which aims at mainly reducing motor symptoms. The advancement of stimulation protocols for DBS neurosurgery has enabled access to the electrophysiology of deep brain structures related to PD, in particular, the basal ganglia, such as the subthalamic nucleus (STN) and internal globus pallidus (GPi). The present study aims to characterize the electrophysiological signals of patients with PD using classical power spectral density (PSD) estimation methods and more recent strategies based on the parameterization of the power spectrum, exploring its particular electrical characteristics (e.g., profile decay). The main objective is to identify possible biomarkers of motor status (i.e., rest or movement) in patients with PD and elucidate possible mechanisms of the disease, allowing in the future a more precise adjustment of DBS protocols aligned with the individual needs of patients. The results obtained demonstrate that there are differences in the signal attributes between states of rest and movement both in classical frequency analysis and in spectral parameterization, but they occur in different intervals of the beta rhythm present in local field potentials, depending on the approach adopted. In particular, the slope of the aperiodic line, a parameter obtained through spectral parameterization, was the best individual indicator of the motor state when analyzed at the central frequency in the narrow band of 24 to 32 Hz, with the area below the Receiver Operating Characteristic curve (AUROC) of 0.7, demonstrating its potential as a biomarker of the motor status of Parkinson's disease. The combination of attributes, including the slope of the straight line with the classical frequency analysis attribute, increased the classification accuracy to 71%, highlighting the importance of spectral parameterization methods based on the decay profile to discriminate PD motor states. Therefore, an in-depth discussion of the interpretation of these parameters for the analysis of the electrophysiological characteristics of PD can contribute to the search for more precise biomarkers and therapeutic strategies and more personalized and effective DBS protocols.
