Temporal lobe epilepsy (TLE) is a neuronal disorder characterized by recurrent and spontaneous
epileptic seizures that can develop from status epilepticus (SE). Among the characteristics of the disease,
it is observed increased neuronal excitability and synchronization, neuroinflammation, glial cell
recruitment and cell death. Coordination of the neuroinflammatory response mainly involves glia with
toll-like receptors (TLRs). In most TLR signaling pathways, the adapter protein myeloid differentiation
primary response 88 (MyD88) is essential. Intrahippocampal delivery of kainic acid (KA) and
intraperitoneal administration of pentylenetetrazol (PTZ) are two experimental models commonly used to
better understand the pathophysiological processes of TLE. Both models were used to evaluate the role of
MyD88 in TLE. To this end, C57BL/6 Wild Type (WT) and MyD88 knockout (KO) mouse lineages were
employed. Behavioral analyses were performed after SE induction in the acute period, and immunofluorescence was employed to evaluate glial response and cell death in the latent period. MyD88
KO has successfully diminished mortality and lowered severity during SE caused by PTZ administration.
During KA-induced SE, MyD88 KO animals showed a higher diversity in Racine scale seizures.
Surprisingly, cell death has spread in both hemispheres of the MyD88 KO, but not in WT controls. In the
latent period, we observed higher presence of microglia cells in C57BL/6 animals, which is reverted in
the MyD88 KO. Next steps include electrophysiological recordings and proteomic analysis. With this
project, we aim to elucidate the mechanisms by which the MyD88 acts in neuroinflammation triggered by
SE, supporting new therapeutic strategies to treat TLE.
