Temporal lobe epilepsy (TLE) is a neuronal disorder characterized by recurrent and spontaneous epileptic seizures that can develop from status epilepticus (SE). Key characteristics of the disease include increased neuronal excitability, synchronization, neuroinflammation, recruitment of glial cells, and cell death. Coordination of the neuroinflammatory response mainly involves glial cells and toll-like receptors (TLRs) signaling, which includes the adapter protein myeloid differentiation primary response 88 (MyD88). Intrahippocampal delivery of kainic acid (KA) and intraperitoneal administration of pentylenetetrazol (PTZ) are experimental models commonly used to better understand the pathophysiological processes of TLE and acute epileptic seizure susceptibility, respectively. Therefore, we employed C57BL/6 Wild Type (WT) and MyD88 knockout (KO) to investigate the role of MyD88 in both KA and PTZ models. Behavioral analyses were performed after SE induction in the acute period to evaluate the intensity of epileptic seizures in both genotypes. Changes in glial cell morphology and cell death in the latent period were also evaluated. Our results demonstrated that the KO of MyD88 reduces mortality and decreases the severity of SE after administration of PTZ as well as KA. After administration of KA, cell death is observed in both hemispheres. In the latent period, both genotypes have glial presence and similar morphology. Electrophysiological results demonstrated similar basal activity. This project evaluated the extent of protection conferred by MyD88 KO during an acute seizure and investigated possible cellular or electrophysiological changes in the latent period as a basis for this protection.
