LIQUID OPTICAL BIOPSY USING FOURIER TRANSFORM INFRARED ABSORPTION TO IDENTIFY BIOMARKERS OF INFLAMMATION

RESUMO:

In recent years, techniques that offer greater precision and speed in diagnosing and monitoring the progress of treatments have aroused great interest, especially those that are minimally invasive for the patient. Individuals affected by pathologies that are considered global health problems, such as cancer, cardiovascular disease, and other chronic illnesses, can benefit from rapid diagnostic methods, which contribute to the implementation of personalized treatments and improve quality of life. FT-IR spectroscopy provides molecular information through optical phenomena observed by the vibration of its molecules. It has been used in biological studies to characterize molecular alterations in various pathologies. In this work, we propose an investigation of salivary biofluid samples using Fourier transform infrared spectroscopy (FT-IR). Our study includes patients with periodontitis, peri-implantitis, and oral cancer (OSCC), as well as individuals who use electronic cigarettes and crack. The main objective was to identify specific biomolecules that could be used as biomarkers in various inflammatory processes, both those resulting from the aforementioned oral pathologies and those associated with drug use. In addition, we sought to explore possible associations and common inflammatory mechanisms between these different clinical conditions, contributing to a better understanding of their effects. The results showed common vibrational signatures that suggest the activation of interconnected immunometabolic pathways, with emphasis on bands associated with phosphate metabolism (945-960 cm-1) and oxidative stress/lipoperoxidation (1712-1762 cm-1). In addition, discriminating bands allowed differentiation between inflammatory oral diseases (1457, 1500, 1581, 1722-1749 cm-1) and drug user groups (985, 1043, 1313, 1400-1563, 1776, 1795 cm-1), reflecting distinct biochemical profiles related to lipid, protein and extracellular matrix metabolism, as well as different patterns of immune modulation. We conclude that salivary FT-IR represents a promising approach for early diagnosis, clinical monitoring, and understanding the molecular mechanisms underlying different inflammatory conditions, distinguishing not only the presence of an inflammatory state but also its specific origins.
