SUMMARY:

Photodynamic therapy (PDT) is a medical treatment in which a combination of a photosensitizing drug and visible light produces highly cytotoxic reactive oxygen species (ROS), leading to cell death. One of the main disadvantages of PDT for topical treatments is the limited penetration of some commonly used photosensitizers in this therapy. In this research, we developed 3D-printed hydrogels and micro-needles with varied concentrations of silk fibroin derived from silk fibers, used as a matrix to incorporate photosensitive molecules with the aim of improving and facilitating delivery for photodynamic therapy. Rheology, spectrophotometry, and scattering techniques were employed to analyze the properties of the resulting hydrogels and micro-needles, in order to elucidate the underlying factors involved in their formation, as well as to investigate the behavior of silk fibroin after the incorporation of the porphyrin into the matrix.
