IDENTIFICATION OF E. FAECIUM AND E. FAECALIS ISOLATED FROM SAMPLES OF SWINE WASTE TREATED BY ANAEROBIC BIODIGESTION: EVALUATION OF RESISTANCE TO ANTIMICROBIALS USED IN SWINE FACULTURE

SUMMARY:

The use of pig manure as fertilizer is a sustainable practice of using solid waste. In pig farming, the use of antimicrobials favors the selection of resistant microorganisms in the intestinal tract of animals and enables the dissemination of multiresistant variants to other living beings. Enterococcus are present in the intestine of animals and are characterized as one of the bacteria that most cause infections, being a public health concern. Considering the relevance of technical-scientific knowledge on antibiotic resistance, the general objective of this study was to analyze the presence of microorganisms of the genus Enterococcus in swine manure samples, which underwent anaerobic biodigestion treatment. The samples were collected in a pig farm in the municipality of Ibiúna (SP). There were four collections, in 2022, of liquid and solid samples. The experimental tests covered the determination of pH and solids; Gram stain, catalase test, primary bacterial isolation of samples in saline solution (incubated at 37ºC for 24h) and, after, cultivation in azide broth culture media (37ºC for 24h), Brain and Heart Infusion (BHI) agar (37ºC for 24h), selective agar medium for Enterococcus (37ºC for 24h); the antibiogram was performed with amoxicillin disks at two concentrations (2µg and 10µg) in culture media with and without food additives added to the animals' feed. From the microorganism in BHI broth with pH 7.8 buffer solution, the conventional polymerase chain reaction (PCR) molecular test was performed with specific primers for Enterococcus; the PCR product was amplified on a 1.5% agarose gel. The positive control was the strain ATCC 29212. Of the total of 8 samples (4 liquid + 4 solid), the study indicated the presence of Enterococcus in the majority (87.5%); molecular tests confirmed the presence of Enterococcus, even in the same number of samples from microbiological tests. The antibiogram did not show microorganisms resistant to amoxicillin, both in the culture media where food additives were added, as well as in those that did not.
