RESUMO:

Nitric oxide (NO) is related to several biological processes, then presenting diverse applications, as antibacterial action. Silver nanoparticles (AgNPs) are also well known for its antibacterial activity. In this work, AgNPs, biogenically synthesized using green tea (Camellia sinensis) extract, were combined with S-nitrosomercaptosuccinic acid (S-nitroso-MSA), a molecule that produces NO. A natural, biodegradable and biocompatible polymer named alginate was used to produce a hydrogel to concentrate and deliver S-nitroso-MSA and/or AgNPs. The material was characterized and healthy lineage Vero cells were used to evaluate cytotoxicity and to determine a safe concentration of application. Gram-negative bacterium Escherichia coli ATCC 25922, and two gram-positive bacteria: Staphylococcus aureus ATCC 25923 and Streptococcus mutans UA159 were used to evaluate a potential antibacterial action and to investigate a possible synergism of action between NO and AgNPs. Hydrogels containing S-nitroso-MSA showed sustained NO release for 18 h at skin temperature. S-nitroso-MSA and AgNPs containing hydrogels presented antibacterial action at 2 μg/mL hydrogel with significantly higher activity compared to hydrogel containing, individually, S-nitroso-MSA or AgNPs. This comproved a synergistic effect for antibacterial action and the in vitro cytotoxicity on Vero cells showed that concentration 2 μg/mL hydrogel is safe for healthy cells. Therefore, alginate hydrogels do have potential for topical antibacterial applications with synergism of action between NO and AgNPs.
