EVALUATION OF THE ANTIMICROBIAL AND ANTI-Trypanosoma cruzi ACTIVITY OF THE APOPHINENIC ALKALOID DICENTRINE AND QUATERNIZED DERIVATIVES

Chagas disease is a neglected tropical disease caused by the protozoan Trypanosoma cruzi, which affects more than 6 million people in Latin America and leads to more than 12 thousand deaths every year. It is essential to search for new active substances against T.cruzi, since the drugs currently available are toxic and have little effectiveness in the treatment of the chronic stage of the disease. Regarding the search for new antimicrobial substances, this is due to the urgency caused by multidrug-resistant microorganisms, leading to the scarcity of effective treatments against these pathogens. In the context of these two problems, and based on the premise that special metabolites from plants are an important source of bioactive substances, this work seeks to contribute to the comparative study of the anti-T.cruzi and antimicrobial activities of the aporphine alkaloid (S)-dicentrin , obtained from the methanolic extract of O.puberula, and three quaternized derivatives: N-methyl-(S)-dicentrin iodide, (S)-dicentrin-β-N-oxide and (S)-dicentrin-α-N-oxide . The four aporphins were obtained and properly characterized by spectroscopic and spectrometric techniques. Preliminary results of anti-T.cruzi activity were obtained for the N-methylated derivative, which showed an EC50 of 44.3 μM against the amastigote form (IS >4.5) and was inactive against the trypomastigote form. This result is in addition to the previously reported result of (S)-dicentrin-β-N-oxide of natural origin, which has an EC50 of 17.4 μM against the trypomastigote form (IS >11.0) and is inactive against amastigote. Both quaternized derivatives have different impacts on the activity of the original alkaloid (S)-dicentrin (EC50 76.8 μM against trypomastigote, with IS <1.0), suggesting that the nature of the quaternization may be determinant for the compound to show activity. The results of antimicrobial activity of the alkaloids in question are still under evaluation and are not yet reported in this work.
