As the life expectancy of the population increases, the incidence of neurodegenerative diseases increases, and it is more likely to know someone who is affected. In Alzheimer's disease (AD) the main symptom seen in patients is memory loss. A consequence and/or aggravating factor of AD is the deregulation of metallic levels in cells and organs. The objective of this work is to elucidate the ionomic profile by inductively coupled plasma mass spectrometry (ICP-MS) of blood plasma, frontal cortex and hippocampus in the in vivo model with streptozotocin (STZ) only or with the presence of the antioxidant hydroxytyrosol (HT, 4-(2-hydroxyethyl)benzene1,2-diol); and also to demonstrate the effect generated in the Barnes behavioral test also reflects the effect it generated in Barnes maze test. Wistar rats were used with 2 intracerebroventricular (ICV) injections of STZ (3 mg/kg, in citrate buffer) only or preliminary injections of HT (1 µL/450 µM). In the 4th week of treatment, animals were subject to Barnes' maze and then euthanasia, where the plasma, frontal cortex, and hippocampus tissues were collected postsaline perfusion. The collected tissues were analyzed with ICP-MS in isotopes of Ca, Co, Fe, Zn, Mn, and Cu. The Barnes maze showed significant differences in the trial and training, in which the STZ group showed less certainty in exploration in the Barnes maze compared to the control group. The STZ+HT group did not show significant differences, with the results closer to the control group. In the analysis of the ionic profile of the tissues with ICP-MS, the blood plasma of the STZ group showed lower cobalt concentration compared to the control, the STZ+HT group showed equivalence to the control in this and other tested elements of the plasma. In the frontal cortex, the calcium concentration of the STZ+HT group is higher than the control, resulting in an aggravating effect on the concentration of this metal with the use of HT. The copper concentration was higher in the STZ group compared to the control, while the STZ+HT group was equivalent to the control group. In the hippocampus, concentrations of calcium and iron were higher in the STZ group compared to the control; the STZ+HT group was similar to the control. The cobalt showed a lower concentration in STZ+HT group compared to the control group. The results demonstrate slight effectiveness of treatments in the Barnes maze with few significant differences and minimal positive effects of TH. The analysis by ICP-MS demonstrates a change in the ionic profile caused by STZ alone or with HT, similar to that seen in the clinical literature on AD. However, it showed inefficiency when changing Zn and Mn concentrations.
