RESUMO:

The resistance of microorganisms to drugs and the search for adequate treatments for neglected diseases are increasingly significant problems in society. Therefore, new alternatives are needed for the treatment of infections, with greater antimicrobial activities, high selectivity and lower toxicity.
Antimicrobial peptides (AMPs) are a therapeutic alternative promising, as they have diverse applicability and wide versatility in their sequential field, thus acting as a potent antimicrobial.
In the present work, studies were carried out aimed at new PAM designs from different organisms. The first group of peptides originates from the venom of a scorpion, of the species Isometrus maculates, and its original peptide is
Imcroporin, in which the study was focused against Chagas disease. Thesecond study group comes from Balteatide, a peptide isolated from the secretion of frog skin, of the species Phyllomedusa baltea, in which the
experiments were focused on bacteria.
The peptides used in this work were synthesized by the solid phase method, using the F-moc strategy, purified by HPLC and characterized by LC/ESI-MS. In biological studies against Trypanosoma cruzi, the trypanocidal effect was evaluated in epimastigote, trypomastigote and amastigote forms.
Toxicity was evaluated in LLC-MK2 host cells by MTT and the selectivity index (SI) was determined. The mechanism of action in T. cruzi was studied in epimastigote forms by flow cytometry using the markers 7AAD/AX, DCF, Rho 123 and by scanning electron microscopy (SEM). The experiments with bacteria had an inhibition test of microorganisms from the ESKAPE group, the mechanism of action was investigated by membrane permeabilization and depolarization and the infectious action, in vivo, was also performed by means of skin infection by abrasion of mouse.
The results of the Imcroporin peptide family were excellent, with the analogue [K] 3 -Imcroporin-NH 2 as the protagonist, which fulfilled the requirements to be a promising antichagasic. And the results with bacteria showed [K] 1 -Imcroporin-NH 2 as a potent antimicrobial in overcoming microbial resistance. The approach of sequential modification of peptides provides important information between physicochemical properties and antimicrobial effect. So that in the future, this class of molecules will, in fact, be the new chemotherapeutic drugs that society needs.
