Air and Surface SARS-CoV-2 Environmental Assessment in the Surrounding of a Field Hospital

With the implementation of a Field Hospital on the premises of UFABC, development of tools to monitor the occurrence of SARS-CoV-2 contamination of the Hospital surroundings are of interest for planning actions and risk prevention for members of the academic community of the University, and could serve as a monitoring model for others airborne infectious diseases Field Hospitals. For air monitoring a  sampling pump model SKC AirChek TOUCH was used for the collections, operating with a flow of 4 l/min, for a period of 30 minutes each sample, connected to a Button type sampler with a 25 mm gelatin membrane filter. in diameter and during the collection, the analysis of other environmental parameters is also performed such as: air velocity, dry bulb temperature, humidity, air velocity, CO2 level, effective temperature and temperature according to the AccuWeater website to correlate in case of positive result. Surfaces evaluation was performed using insects as sentinels. Ten ants and two hind legs of a cockroach were collected in the gardens next to the Field Hospital building. After the collections, the gelatin membrane and the insects were used for nucleic acid extraction and detection of SARS-CoV-2 by reverse transcription (RT) and quantitative polymerase chain reaction (qPCR) in real time and also endpoint PCR. RTqPCR allows the detection of virus RNA and was performed in the pathogenic agents laboratory at UFABC. Endpoint PCR was used for detection of SARS-CoV-2 Envelope and Spike fragments encoding genes from insect nucleic acid and amplicons of positive samples were cloned and sequenced by Sanger’s method. One ant and one cockroach leg were positive for the E and Spike fragments, respectively, and the SARS-CoV-2 specific sequences were confirmed by sequencing. In addition to the detection of SARS-CoV-2 RNA, it was evaluated the presence of human genetic material in the gelatin filters and in insects, where one gelatin filter presented positive result.
