Cardiorenal syndrome (CRS) is a pathological process involving the kidneys and heart in such a way that dysfunction in one organ can trigger damage to the other organ. CRS 3 is characterized by acute kidney injury as a precursor to subsequent cardiovascular changes. Previous studies have shown the participation of reactive oxygen species (ROS), such as nitric oxide (NO) and superoxide anion (O2-) in SCR3. It is known that mitochondria are dynamic organelles that respond to different stimuli, through the processes of mitochondrial fission and fusion. Also, they are the main sources of production of EROs. It is known that vitamin C, an exogenous antioxidant, has a proven protective capacity for cardiac, renal and vascular endothelial tissues. Given that, the present study aimed to evaluate the function and mitochondrial dynamics in renal and cardiac tissues during SCR3, as well as the role of vitamin C in this process. The unilateral renal ischemia (I / R) model was performed for 60 minutes, followed by reperfusion for 24h, 48h, 72h, 8 days, 15 days and 23 days, with and without vitamin C treatment, immediately after I / R or 15 days after I / R. Renal and cardiac morphometric parameters, biochemical parameters (urea and creatinine) and molecular markers were evaluated through gene expression. Mitochondrial function was assessed by high resolution respirometry and mitochondrial dynamics through the expression of OPA1 and FIS1. NO levels were assessed using the amperimetry technique. In the renal tissue, a reduction in mass, an increase in urea levels, a reduction in oxygen consumption, an increase in NO levels and a reduction in OPA1 gene expression were observed after 15 days of I / R. Treatment with vitamin C preserved renal mass, restored function and reduced NO levels with late treatment. Cardiac tissue showed an increase in ANF expression, reduced oxygen consumption and decreased NO levels. Under treatment with vitamin C immediately after I / R, NO levels were restored. In conclusion, the results indicate that I / R induced impairment in mitochondrial function and dynamics in the kidneys and heart. Still, treatment with vitamin C was able to promote a renoprotective effect.
