Preparation, characterization and biological assays of polymeric nitric oxide donors biomaterials

PÁGINAS: 66

ÁREA: Química
RESUMO:

Nitric oxide (NO) is a small molecule, lipophilic and devoid of
which has a myriad of biomedical applications, such as antitumor activity,
antibacterial activity and vasodilation. One of the disadvantages for the clinical use of NO is its
relatively short half-life of 1-5 s. To make its therapeutic application viable, the
of this work was to incorporate NO-donor molecules, specifically S-nitrosothiols (RSNO)
in chitosan nanoparticles (CS NPs) and hydrogels. The CS NPs were synthesized by means of
ionotropic gelation using sodium tripolyphosphate (TPP) as a counter ion,
CS NPs were obtained containing the donor molecule NO mercaptosuccínico Snitroso acid (MSA Snitroso-CS-NPs), with an average hydrodynamic size of 142.80 ± 2.22 nm. after the
with the hyaluronic acid (HA), the mean hydrodynamic size increased to
170.80 ± 0.14 nm. Cytotoxicity assays demonstrated a toxic effect on
(PC3 and HeLa) with a pronounced toxic effect on the nanoparticles coated with
AH. In addition to the application of CS NPs to tumor cells, the molecule
of S-nitrosoglutathione (GSNO) therein for application to human skin. The GSNO is a
endogenous molecule belonging to the group of RSNOs. When applying the GSNO-CS NPs in the
epidermis of human samples, an increase in the concentration of S-nitrosothiols
(RSNOs) in 32% of the skin, indicating that GSNO-CS NPs can release NO and the
even penetrates the human skin causing the dermal levels of RSNO to increase. In addition to
nanoparticles containing NO donors, the potential of
chitosan (CS-Hid) containing NO donors. In the formulation of the hydrogel tested, Pluronic F-127 was added, which gives the material thermoresponsive behavior. Reviews
rheology and morphology demonstrated a hydrogel with gelation temperature (Tgel) of
29.95 ± 0.85 ° C and good mechanical properties. Cytotoxicity assays have demonstrated
concentrations to be administered in mammalian cells with potent
bactericidal effects (Pseudomonas aeruginosa). Two materials were evaluated
donors with potential for biomedical applications,
bactericidal and antitumor action.
