RESUMO:

Autism Spectrum Disorders (ASD) are a group of neurodevelopmental disorders which present wide genetic heterogeneity, being considered a complex disease depending not only on innumerable genetic factors, but also on environmental factors. Unraveling the genetic architecture responsible for the pathogenicity of this disorder is considered an open problem. High-performance sequencing technologies made possible the acquisition of large amounts of genome data, gene expression, and proteomes of individuals with this disorder. The integration of these data based on the hypotheses of network medicine may help in the prioritization of genes potentially associated to this disorder. Recently a method called NERI (Network Medicine Relative Importance) for gene prioritization has been developed which models the hypotheses of network medicine for gene prioritization based on integration of gene expression, genome association studies (GWAS) and protein-protein interaction networks (PPI). The potential of this method to recover relevant genes and discover new genes associated with a complex disease was demonstrated through results of prioritization of genes for schizophrenia. The objective of this project is to analyze the NERI method by including copy number variations (CNVs) and exome, since the literature has shown that these types of data are very relevant in the characterization of neurodevelopmental disorders as ASD. We hope that the results of this project will provide inputs to improve the NERI method to include CNV and exome data effectively in its integrative model, allowing a larger efficacy in the search for ASD biomarkers.
