RESUMO:

Autism spectrum disorder presents large phenotypic and genotypic heterogeneity, being characterized as a complex disease. The genetic architecture of this disorder is composed of rare inherited and de novo variants. The genes impacted by these variants converge on biological pathways. The topology of interactions among these genes and with phenotypes at the individual level is poorly explored. We investigated whether it is possible to group patients from their topologies of integrated biological networks. With this aim, we selected TEA subgroups based on components of exposure scores for environmental and biological risk factors and compared the topologies of the prioritized subnetworks. Patients had their exomes sequenced from which we selected rare variants and de novo variants, mapped the variants to the human interatoma, and prioritized interactions based on each gene's topology and prefrontal cortex expression pattern. From the prioritized genes and interactions of each patient and the integration with co-expression data through the NERI method, we characterized the topology with 40 local topological measures and the global topology with measures from the information theory. The study of subnetwork topologies in developmental disorders has the potential to reveal emerging features useful for stratification of subgroups of patients.
